research.<\/em><\/p><p>The FDA recently <strong>approved the immunotherapy drug pembrolizumab<\/strong> as a first-line treatment for patients with certain types of <strong>advanced colorectal cancer<\/strong>. This is the 9th FDA approval supported by Stand Up To Cancer\u00ae (SU2C) research.<\/p><p>Patients newly diagnosed with <strong>advanced or metastatic microsatellite instability-high (MSI-H)<\/strong> or <strong>mismatch repair deficient (dMMR) colorectal cancer<\/strong> previously would have been prescribed pembrolizumab only after exhausting standard chemotherapy treatments.<\/p><p>The FDA approved pembrolizumab, also known under the brand name <strong>Keytruda<\/strong>, as first-line treatment for <strong>metastatic MSI-H\u2014dMMR colorectal cancer<\/strong> based on early results from a phase III clinical trial, which was partially funded by an SU2C grant.<\/p><p>\u201cWhen compared to traditional treatment, pembrolizumab was superior with fewer side effects for MSI-H\u2014dMMR colorectal patients,\u201d said Luis A. Diaz, MD, Head of the Division of Solid Tumor Oncology at Memorial Sloan Kettering Cancer Center and leader of the\u00a0<a href=\"https:\/\/standuptocancer.org\/research\/research-portfolio\/dream-teams\/colorectal-cancer-vulnerabilities-dream-team\/\">SU2C Colorectal Cancer Dream Team<\/a>, which conducted the research. \u201cThere are other solid tumor cancers in adults and children that have these same MSI-H\u2014dMMR defects, so our research may also have ramifications for other cancer types.\u201d<\/p><p>Around 4-5% of metastatic cancer tumors have MSI-H\u2014dMMR biomarkers, which result from the inability of cells to repair mistakes made during the cellular division process and can lead to more tumor development.<\/p><p>The study involved 307 MSI-H\u2014dMMR colorectal cancer patients in 23 countries who were treated either with pembrolizumab or standard chemotherapy. Pembrolizumab targets and blocks a protein called PD-1 that can prevent immune cells called T cells from eliminating cancer cells effectively.<\/p><p>Dr. Diaz and his team found that MSI-H\u2014dMMR colorectal cancer patients treated with pembrolizumab didn\u2019t see their cancer spread for a median 16.5 months, compared to patients treated with standard chemotherapy who saw their tumors grow after a median 8.2 months. Patients receiving standard chemotherapy also had more severe side effects than patients receiving pembrolizumab. The complete study results\u00a0<a href=\"https:\/\/www.nejm.org\/doi\/full\/10.1056\/NEJMoa2017699?af=R&amp;rss=currentIssue\" target=\"_blank\" rel=\"noopener\">were published<\/a>\u00a0in the New England Journal of Medicine on December 2, 2020.<\/p><p>\u201cThe Stand Up To Cancer Colorectal Cancer Dream Team has made a significant contribution to an improved treatment option for MSI-H\u2014dMMR colorectal patients,\u201d said Nobel laureate Phillip A. Sharp, PhD, chair of Stand Up To Cancer\u2019s Scientific Advisory Committee and an Institute professor at the David H. Koch Institute for Integrative Cancer Research at Massachusetts Institute of Technology. \u201cThis is a great example of how Stand Up To Cancer\u2019s collaborative research model has a direct impact on cancer patients\u2019 lives.\u201d<\/p><p>Colorectal cancer is the\u00a0<a href=\"https:\/\/www.cancer.org\/cancer\/colon-rectal-cancer\/about\/key-statistics.ht\" target=\"_blank\" rel=\"noopener\">second most common cause of cancer death<\/a>\u00a0among American men and women combined and nearly\u00a0<a href=\"https:\/\/www.cancer.org\/content\/dam\/cancer-org\/research\/cancer-facts-and-statistics\/colorectal-cancer-facts-and-figures\/colorectal-cancer-facts-and-figures-2020-2022.pdf\" target=\"_blank\" rel=\"noopener\">148,000 Americans<\/a>\u00a0will receive a new diagnosis of colon or rectal cancer in 2020. While colorectal cancer death rates have declined significantly due to increased screening and improvements in treatment, about\u00a0<a href=\"https:\/\/www.cdc.gov\/media\/releases\/2013\/p1105-colorectal-cancer-screening.html#:~:text=About%20one%20in%20three%20adults,for%20Disease%20Control%20and%20Prevention.\" target=\"_blank\" rel=\"noopener\">1 in 3 adults age 50 or older<\/a>\u00a0do not get the recommended screening. New cases of colorectal cancer are\u00a0<a href=\"https:\/\/www.cancer.org\/latest-news\/study-finds-sharp-rise-in-colon-cancer-and-rectal-cancer-rates-among-young-adults.html\" target=\"_blank\" rel=\"noopener\">occurring at a growing rate<\/a>\u00a0among young and middle-aged adults in the US, with the number of cases of colorectal cancer\u00a0<a href=\"https:\/\/www.mskcc.org\/clinical-updates\/msk-s-new-center-focus-alarming-increase-colorectal-cancer-young\" target=\"_blank\" rel=\"noopener\">in people under 50 expected to almost double<\/a>\u00a0by 2030.<\/p><p>As colorectal cancer continues to disproportionately impact people of color<br \/>(<a href=\"https:\/\/www.cancer.org\/content\/dam\/cancer-org\/research\/cancer-facts-and-statistics\/cancer-facts-and-figures-for-african-americans\/cancer-facts-and-figures-for-african-americans-2019-2021.pdf\" target=\"_blank\" rel=\"noopener\">Black people have the highest rates<\/a>\u00a0of colorectal cancer of any racial or ethnic group in the US), improvements in screening and new precision and targeted treatments must reach all patients. SU2C announced a Health Equity Initiative in January 2020. The initiative requires all future teams seeking Stand Up To Cancer funding to address recruitment and retention of patients from different ethnic and racial groups and underserved communities to improve diverse participation in cancer clinical trials.
